"Revisiting an ancient histone variant in an emerging insect model: the cricket Gryllus bimaculatus"
Résumé :
We are also developing the cricket Gryllus bimaculatus as an emerging representative of hemimetabolous insects. Among many points of interest, the Gryllus genome is rich in histone variant genes that are not represented in holometabolous model species, including Drosophila. Here, I will present our functional characterization of the single Gryllus macroH2A gene. MacroH2A is the most atypical histone variant described so far, being three times larger than canonical H2A due to the presence of a 25kDa macrodomain at its C-terminus. Despite efforts to unravel its role in vertebrate models, the molecular function of this ancient histone remains poorly understood. Our study reveals a remarkable association of macroH2A in the chromatin of differentiating cells in developing embryo. We also demonstrate that the role of macroH2A in preserving genome integrity is conserved from insects to mammals.
